Key Responsibilities
- Isolation, culture and lentiviral transduction of primary cells using aseptic techniques.
- Perform cellular and molecular assays (e.g. flow cytometry, cell sorting, PCR, qPCR, ddPCR, LAM-PCR, western blot).
- Generate molecular libraries for RNA-seq, scRNA-seq, ATAC-Seq, CITE-seq and other high throughput techniques and run Illumina sequencing.
- Perform exploratory analyses on biological samples from patients enrolled in AVROBIO’s clinical trials using immunophenotyping, molecular and single cell sequencing assays.
- Support animal experiments locally and at CROs.
- Potentially support the R&D team cross-functionally, i.e. other programs and assist in the training of new technical staff.
- Maintain an electronic laboratory notebook that provides clear documentation of experimental methods and results.
- Work with the laboratory manager to maintain equipment and supplies.
Minimum Requirements/ Qualifications
- BS or MS degree with 2-4 years of laboratory experience (industry experience will be considered a plus).
- Solid base of cell culture and aseptic techniques (experience with human primary cells will be considered a plus).
- Experience with multicolor flow cytometry (experience with FACS cell sorters will be considered a plus).
- Extensive experience with molecular biology techniques including DNA/RNA isolation, cloning, PCR and RT-PCR (experience with library preparation for high-throughput sequencing will be considered a plus).
- Experience with mouse work preferred, but not required.
- Ability to troubleshoot and optimize protocols with the assistance of a supervisor.
- Ability to quickly learn new techniques and adapt to a fast pace environment.
- Must demonstrate careful attention to detail alongside excellent organizational, time management, and communication skills.
- Must enjoy working as part of a cross-functional team and have interest in the field of gene therapy.
